Sulfo-Cy5 NHS ester (potassium) is a water-soluble reactive dye used for the labelling of proteins and nucleic acids. Sulphated Cy5-NHS ester is highly water-soluble; consequently, no organic co-solvents are required during the labelling reaction, making it particularly suitable for labelling biomolecules such as proteins that are sensitive to organic solvents. Furthermore, sulphated Cy5 is highly water-soluble and the dye molecule itself is charged; once labelled onto the protein surface, it does not induce aggregation of hydrophobic proteins, thereby enhancing the stability of the fluorescently labelled product.
Important notes
The NHS ester group is sensitive to humidity; therefore, the product must be stored in a dry environment. Allow the product to equilibrate at room temperature for at least 20 minutes prior to use.
Use a buffer system that does not contain amine groups. Examples include sodium phosphate buffer (PBS), HEPES buffer, sodium bicarbonate buffer, or borate buffer. Do not use any buffers containing primary or secondary amines, such as Tris or glycine. High concentrations of imidazole may also affect the reaction.
Do not expose the labelling reaction to strong light, as most fluorescent compounds undergo gradual photobleaching when exposed to strong light for prolonged periods.
