Biological Activity

IL-27 is a heterodimeric cytokine that belongs to the IL-6/IL-12 family of long-type type I cytokines. It is expressed on monocytes, endothelial cells and dendritic cells. IL-27 enhances the early phase of the TH1 response and inhibits the differentiation of TH2 and TH17. It induces TH1 cell differentiation through two distinct pathways, the p38 MAPK/TBX21 and the ICAM1/ITGAL/ERK-dependent pathways. It also induces phosphorylation of STAT1, STAT3, STAT4 and STAT5 and activates TBX21/T-Bet via STAT1, resulting in upregulation of IL12RB2, an event critical for TH1 cell commitment. IL-27 has anti-proliferative activity against melanoma via WSX-1/STAT1 signaling.
