Biological Activity

Interleukin-17 is a potent pro-inflammatory cytokine produced by activated memory T cells. There are at least six members of the IL-17 family in humans and mice. Because IL-17 has the same properties as IL-1 and tnf-α, it may cause joint inflammation and bone and cartilage destruction. This cytokine is present in the synovial fluid of patients with rheumatoid arthritis and is produced by the synovial membrane of rheumatoid arthritis. It increases IL-6 production, induces collagen degradation, reduces collagen synthesis in synovium and cartilage, and proteoglycan synthesis in cartilage. IL-17 also increases bone destruction and reduces its formation. Blocking interleukin-17 with specific inhibitors provides protective inhibition of cartilage and bone degradation.

Measured by its ability to induce IL-6 secretion by NIH‑3T3 mouse embryonic fibroblast cells, the ED50 for this effect is 1-7 ng/ml.

Recombinant human CXCL13 is a single non-glycosylated polypeptide chain containing 87 amino acids and mature human BCA-1 shares 64 % amino acid sequence similarity with the murine protein and 23–34 % amino acid sequence identity with other known CXC chemokines. Determined by a chemotaxis bioassay using human B cells, the biological activity is in a concentration range of 1.0-10 ng/ml.
