Biological Activity

PKH-26 is a red fluorescent dye, PKH-26 can stably bind to the lipid region of cell membrane and emit red fluorescence (Ex/Em=551/567 nm), which is mainly used for in vitro cell labeling, in vitro cell proliferation studies and in vivo and in vitro cell tracing studies.


PKH-26 Exosome Staining Protocol (for reference only):
A PKH-26 kit was employed to label Exo according to the manufacturer's protocol. Typically, the Exo was incubated with 100 μM PKH-26 dye solution (50 μL of PKH-26 dye solution per 100 μg of Exo) at 37 °C for 10 min in a dark environment. Subsequently, 20 mL of PBS was added to stop the staining. The PKH-26-labeled Exo was obtained through differential centrifugation and resuspended with 500 μL of PBS.

Preparation method The cells are then resuspended in a loading buffer and incubated for 40 min with freshly prepared PKH-26 (stock 1 mM in ethanol) at room temperature. For the loading of tumor cells, it is recommended to use 0.8 μl, for small cells 0.5 μl PKH-26 in 200 μl loading buffer.
