OVA Peptide(257-264) is a class I (Kb)-restricted peptide epitope of OVA, an octameric peptide can be from ovalbumin presented by the class I MHC molecule, H-2Kb.

| Preparation method | Mice were subcutaneously immunized with 50 µl of 2 mM KFE8 nanofibers (without epitope), 2 mM OVA nanofibers, 2 mM OVA.Tat12.5 nanofibers, OVA protein (25 µg per mouse), or PBS as control. After 1 week, spleens were isolated and single-cell suspensions were prepared, washed three times with PBS, plated into 96-well plates (5 × 10^5 cells in 200 µl/well) and stimulated with 5 μM of soluble OVA257–264 peptide and monensin. Some wells were treated with 5 μM of E749–57 peptide as a nonrelated peptide control. After culture at 37°C/5% CO2 for 6 hr, FC receptors were blocked by addition of anti-CD16/32 antibodies for 30 min on ice, and the cells were stained using APC-conjugated antimouse CD3 antibody and FITC-conjugated antimouse CD8 antibody for 15 min at 4°C. |
