NSC687852 is a cell-permeable, potent, reversible and selective inhibitor of 19S regulatory-particle-associated deubiquitinases (DUBs) UCHL5 and USP14, with no effect on UCH-L1, UCH-L3, USP2, USP7, USP8 and BAP1 and on the proteasomal proteolytic activities. NSC687852 induces cell-cycle arrest and apoptosis in several colon cancers and in CNS cells, and suppresses tumor growth in various mouse models.
