MC4171 is a first-in-class selective KAT8 inhibitor (IC50=8.1 µM). MC4171 exhibits moderate micromolar antiproliferative activity in different cancer cell lines, including NSCLC and AML, without impacting the viability of nontransformed cells. MC4171 (compound 34) exhibited great selectivity over a panel of KATs and KDACs, also displaying the capability to target KAT8 and to exert cancer-selective antiproliferative effects in cells.
