FK962 is an enhancer of somatostatin release, exerts cognitive-enhancing actions. FK962 significantly reduces somatostatin-induced inhibition of Ca2+ channels at 1-100 nM in single rat hippocampal neurons using whole-cell patch-clamp. FK962 (1 nM-1 μM) also significantly enhances high K+-evoked somatostatin release from rat hippocampal slices.
In vivo, FK962 (0.1, 0.3 or 1.0 mg/kg; i.p.) and Donepezil (a single dose of 1 mg/kg) act synergistically to improve cognition in rats.
