Biological Activity

Euphol is a tetracyclic triterpene alcohol isolated from the sap of Euphorbia tirucalli with anti-mutagenic, anti-inflammatory and immunomodulatory effects, orally active. Euphol inhibits the monoacylglycerol lipase (MGL) activity via a reversible mechanism (IC50=315 nM).

Euphol (0.01-0.3 mM; 24-72 hours) markedly inhibits T47D cells proliferation, the IC50 values of euphol treatment for 24, 48 and 72 h were 0.26, 0.22 and 0.13 mM, respectively. Euphol (0.03 mM; 48 or 72 hours) leads to cell cycle arrest by regulating expression of cell cycle-associated proteins.

Preparation method The proliferation effect of euphol was assessed by BrdU assay kit following the manufacturer's instructions. In brief, 5×10^3 cells/well were seeded in a 96-well, flat-bottom plate. The plate was incubated overnight for optimal attachment of adherent lines, and then placed under low serum-starved conditions for 24 h (DMEM-0.5% FBS). Subsequently, the cells were treated with increasing concentrations of the test compound diluted in DMEM (0.5% FBS) and incubated for 72 h. The control groups received the same amount of vehicle (1% DMSO, final concentration). Therefore, the proliferation effect was assessed by BrdU assay kit following the manufacturer's instructions. Experiments were done in triplicate in three independent experiments for eachcell line.
