Biological Activity

Andrographolide is a NF-κB inhibitor that inhibits NF-κB activation by covalently modifying the cysteine residue of P50 in endothelial cells without affecting IκBα degradation or p50/ P65 nuclear translocation. Andrographolide has antiviral effects.

Cell lines MLE-12 cells
Preparation method MLE-12 cells, a cell line derived from murine alveolar epithelial cells, were plated at a density of 5×103 cells per well in a 96-well plate and grown to 80% confluency in Hites media 10% fetal bovine serum. Then, cells were pretreated with 50 µM andrographolide or vehicle (0.01% DMSO) 4 h before LPS (0.5 µg/ml) stimulation. Total and nuclear proteins and mRNA were extracted from cells at specified time intervals. MTT assay was used to assess cell viability.
